Figure Lengend Snippet: Illustration of 3D-SMART integrated with brightfield imaging. A 488-nm laser is used for tracking excitation. It is deflected by a pair of electro-optical deflectors (EODs) and a tunable acoustic gradient (TAG) lens to form a 3D scanning volume. The beam is reflected into the objective by a multiband dichroic mirror (DCM1). A 780 nm LED beam is used for the brightfield imaging excitation and shines directly from the top of the sample. The combined imaging and tracking emission signal were then sepreated by a dichroic mirror (DCM2). The imaging signal was detected by a sCMOS camera and the tracking signal was detected by a single-photon counting avalanche photodiode (APD).
